WiFi Access: Both the kindle and nook have wifi access so you can access the Internet to download books while on the go. The Sony reader doesn't have this yet, you have to access a computer to buy and download a book.
The Amazon kindle has the best Internet access as it also has a browser so you can access the Internet at home and abroad. This means you can access the Internet wherever there's wifi without paying any charges. The Nook's internet access is restricted to download books and only available in the US.
Storage : The Kindle and Nook readers have storage space for storing about 1500 books. The Sony has storage space for about 350 books. However the nook and some models of the sony reader comes with an extra memory card so you can take all your ebooks with you when traveling quite easily. The kindle has no extra memory card but you can back up all your books on your kindle book library at amazon. These can be downloaded wirelessly at any time (of course you'll need to be in an area with wifi access to be able to do this)
Cost and access to e-books : The amazon kindle probably has the access to the most ebooks at the cheapest prices. This is worth taking into consideration as if you're buying a lot of books you will want to buy them at the cheapest prices otherwise it'll be quite costly in the long run. The sony and nook both have their own bookstores. Before you buy it may be worth checking out the books you can buy at each of the reader's stores to see what's available.
Multiple Formats: The sony reader supports multiple formats including Adobe® PDF, Microsoft® Word, image and music files so you can access and read any documents you may have. This will be a big advantage for the sony reader if perhaps you are a student who will need to read word and pdf documents. The kindle reads pdf files but not word format. I think you can send any files you can't read to amazon to convert them for you, but obviously this is a hassle and not something you'd probably want to do a lot.
Touch Screens : The Sony touch screen reader has a touch screen which a lot of people like. You can turn pages by swiping your finger along the screen while you press buttons with the Nook and the Kindle.

I bought the Sony Pocket reader, partly because it's so light. This makes reading more enjoyable. Also, I wanted to take ebooks out of the library. I couldn't do that with a Kindle.
I love paper books, so I was reluctant to buy an ereader, but once I did I knew that ereaders are here to stay. It's just so much easier to hold one than a hardcover book.

Kindle, with all it's advantages, does not allow you to take ebooks from the public library. Sony and Nook do.
I have the Nook and you can browse the internet with it. It does not have flash player so there isn't any videos or gaming to be done, but I access facebook and google with no issues at all.
I have the color Nook and I can access the internet and do almost everything on the internet I do with my home computer. Although it does not have a flash player I can see You Tube videos. I love it , plus it has the droid operating system so I can access app features. It's a great e-reader and more..
